I want to buy a spare that actually matches the wheels on my 2020 Colorado. Can I get the factory wheel size and style out of the RPO codes, or do I need to measure the ones on the truck?

Wheels have their own codes and the description generally covers the size and style. That gets you a matching part number without measuring.

If the truck is on aftermarket wheels now, the code tells you what it came with, which may not be what you need for a spare that fits in the well.

Just confirm the offset when it arrives. Same diameter does not always mean the same wheel.
